In general, gram-negative bacteria have a separate structure called lipopolysaccharide （LPS） in addition to phodpholipid and protein in the external layer of cell envelope, which has its different characteristics depending upon bacteria. Such LPS has been known to not only play an important role in the growth or survival of bacteria but play many roles, particularly in the interaction between host and parasite. In addition, LPS is understood to have a close relation to the immunity reaction, for example it gets endotoxin secreted when septic shock is given and as a result, it brings about peripheral vascular collapse and further LPS is important in the toxic pathophysical reaction. Like this, LPS has a different extraction pattern depending upon bacteria and this enables to separate bacteria systematologically as well. As mentioned earlier, bacterial endotoxin is a material which is generally present in the outer wall of cells in Gram-negative bacteria, which doesn’t appear while bacteria is alive but, when bacteria multiply or dies out, is released out the cell walls breaking. Entotoxin in the state of nature comprises LPS, Proteins and Phospholipids, which is very stable, heat-resistant, takes on negative charge and has bulky molecular weight （1,000,000 dalton or more）. In general, protein is obtained from being extracted with Trichloroacetic acid, Butanol or EDTA and removing lipid associated proteins （LAP）, and phospholipids is extracted using phenol. When we usually attempt to extract LPS, LPS of about 1~4% per bacterial dry weight can be obtained.
LPS Extraction Kit is based on phenol water method and so this kit denatures the protein in entotoxin and then removes through the denaturalization in the form of precipitate between water layer and phenol layer, in addition, this kit separates lipid by way of fraction in Phenol/chlorothe form and as a result, can extract solely intensive LPS quickly and easily.
